Melatonin, a hormone produced by the pineal gland, has been found to be safe for short-term use and may even have a protective effect on the liver in certain situations. It is generally safe for short-term use and does not cause dependence or hangover effects. The most common side effects of melatonin include maintaining liver function with decreased levels of ALT, AST, and alkaline phosphatase, as well as increasing antioxidative functions.

This study is the first systematic review and meta-analysis investigating the effect of melatonin supplementation on liver enzymes. The research program included 60 patients aged 47-65. Several randomized clinical trials (RCTs) evaluated the effect of melatonin supplementation on liver enzymes in patients with non-alcoholic fatty liver disease (NAFLD). Melatonin has not been implicated in causing serum enzyme elevations or clinically apparent liver injury. Melatonin contains antioxidant characteristics, which improve liver tissue injuries and liver enzymes by scavenging endoplasmic reticulum stress.

Hepatotoxicity is a common cause of chronic inflammation, which plays a key role in the pathophysiology of numerous common diseases. Melatonin supplementation can improve liver indices, but more RCTs are required with larger sample sizes and better control. Melatonin significantly decreases liver enzymes, so the use of melatonin in patients with NAFLD can be effective.

Melatonin has also been found to have an improvement effect on blood pressure, decrease the level of elevated liver enzymes, and help weight reduction. A systematic review and meta-analysis included five studies that investigated the effect of melatonin on liver enzymes in 260 participants with NAFLD. Results showed that after 6 months of melatonin treatment, liver enzyme levels were decreased (nearly a 40 decrease in comparison with baseline values, 25 normalized).

In conclusion, melatonin has shown potential benefits in treating NAFLD and other liver diseases, including weight loss and improved liver function. Further research is needed to confirm its effectiveness in treating NAFLD and other liver disorders.

In conclusion, melatonin administration attenuates oxidative stress, inflammation, and restores renal function and structure in rats. If proven effective, melatonin would be an attractive adjunctive therapy due to its natural, inexpensive, widely available, orally administered, and relatively safe product.

Melatonin is used as a marker of the phase of the internal circadian clock and is used as a therapy for certain sleep disorders related to circadian rhythm abnormalities.

The pineal gland’s function and melatonin secretion can be impaired due to accidental and developmental conditions such as pineal tumors, craniopharyngiomas, injuries affecting the sympathetic innervation of the pineal gland, and rare congenital disorders that alter melatonin secretion. The pineal gland is a small, highly vascularized, and secretory neuroendocrine organ located in the mid-line of the brain.
