The brush border, a complex and highly plastic organelle in the small intestine, is responsible for the absorption of nutrients and maintaining intestinal homeostasis. It is composed of numerous microvilli that extend from the cell and form the brush border. These enzymes, known as brush border enzymes, take over from there and are essential for breaking down oligosaccharides. The most important brush border enzymes are dextrinase and glucoamylase, which further break down oligosaccharides.

Brush border enzymes with transmembrane peptide anchors can be cleaved from the membrane by proteolysis, while both peptide and GPI-anchored enzymes are also present. The brush border contains several key enzymes that digest luminal digestion products to produce monosaccharides, including sucrase-isomaltase, lactase, and maltase. In vivo, intestinal epithelial cells are covered by apical membrane structures known as microvilli that constitute the brush border and act as the anchorage site of several specific enzymes.

The apical membrane of proximal tubular epithelial cells contains numerous microvilli that form the brush border. These enzymes carry out the specialized functions of the proximal tubule, such as sucrase-isomaltase, lactase, maltase-glucoamylase, and trehalase. Sucrase-isomaltase is a single gene product with two active sites.

Several lipases are present in the brush border of the small intestine, including phospholipase B1, neutral ceramidase, and alkaline sphingomyelinase. These enzymes are produced by enterocytes that line the intestinal walls and form the brush border.

Intestinal brush border enzymes and pancreatic enzymes are responsible for the majority of chemical digestion, as well as the breakdown of fat. Disaccharidases and alkaline phosphatase are predominantly found in the brush border, and significant amounts of ATPase are also located at this region.
