Ascorbate peroxidase (APX or APEX) is an enzyme that catalyzes the chemical reaction L-ascorbate + H2O2 dehydroascorbate + 2 H2O. It belongs to the family of heme-containing peroxidases and functions as an electron microscopy tag and promiscuous labeling enzyme for live-cell proteomics. APEX is an engineered peroxidase that generates a short-lived biotin-phenoxy radical that nonspecifically labels. This method generates a reaction product within well-preserved, tightly cross-linked cells, unlike the immunoperoxidase method, which creates a permeabilized cellular ultrastructure.

There are two major classes of enzymes currently used for Proton Deposition Mass Spectroscopy (PDB-MS): biotin protein ligases and peroxidases. APEX is used for in-cell biotinylation, catalyzing the oxidation of biotin-phenol to the short-lived biotin-phenoxyl radical in the presence of hydrogen peroxide. Enzyme-catalyzed proximity labeling (PL) is an alternative to capture the close neighborhood of proteins, relying on the targeting of a “promiscuous” tagging enzyme (APEX or BirA) to the region of interest.

Each enzyme binds to one specific substrate, and APEXzymes efficiently uses abundant agro-industrial by-products as raw materials, drastically reducing enzyme production costs. Apexzymes, a startup based in Campinas, São Paulo state, is developing technology to enable enzymes to be produced locally.

  • Bloodwork is perfectly fine however, gaging many vitamins and minerals and other substances through bloodwork can be a matter of inaccuracy, considering the simple fact that many of these substances don’t reside in the blood, but rather in the cells. So any reading of these substances via blood is simply a snapshot of what’s happening in your blood at that very moment in time. Intracellular tests are much more indicative of levels but also much more inaccessible and costly. Just get everything your body needs, and we’ll all be fine.

  • Simply adding supplements is not necessarily going to improve health. For example recent studies show an association with new onset atrial fibrillation(a high risk for new stroke) is associated with taking omega 3 supplements. Don’t listen to nonphysicians for supplements. Always speak to your physician before starting any new supplement.
