A new enzymatic transformation for l-tyrosine synthesis is introduced by demonstrating that the β-subunit of tryptophan synthase natively couples indole and l-Trp. An engineered IGPS enzyme may be able to catalyze the synthesis of new indole derivatives or improve the economical feasibility of indole production through the use of more temperate materials.

Plants overcome this limitation and produce indole by the activity of a different enzyme, indole-3-glycerolphosphate lyase, which forms indole.
