The oxidative breakdown of glucose and fatty acids is the principal source of metabolic energy in animal cells. The initial stages of glucose metabolism (glycolysis) occur in the cytosol, where glucose is converted to pyruvate. Pyruvate is transported into mitochondria, where its complete oxidation to CO 2 yields the bulk of usable energy (ATP). This involves the initial oxidation of pyruvate to acetyl CoA, which is then broken down to CO 2 via the citric acid cycle. The enzymes of the citric acid cycle, located in the matrix of mitochondria, are central players in the oxidative breakdown of both carbohydrates and fatty acids.

The oxidation of acetyl CoA to CO 2 is coupled to the reduction of NAD+ and FAD to NADH and FADH 2, respectively. Most of the energy derived from oxidative metabolism is produced by the process of oxidative phosphorylation, which takes place in the inner mitochondrial membrane. Glycosomes are peroxisome-related organelles found in kinetoplastids and diplonemids, containing enzymes of the glycolytic and gluconeogenic pathways. Biochemical studies and proteome analysis of Trypanosoma cruzi glycosomes have identified enzymes of several other metabolic processes in the organelles. These analyses reveal similarities with glycosomes from other trypanosomatids and features specific to T. cruzi. The enzyme content indicates that T. cruzi glycosomes are multifunctional organelles involved in both catabolic processes such as glycolysis and anabolic ones. The cross-talk between glycosomal metabolism and metabolic processes occurring in other cell compartments is discussed, as well as the importance of metabolite translocation systems in the glycosomal membrane to coordinate spatially separated processes. Possible mechanisms for metabolite translocation across the membrane are suggested by proteins identified in the organelle’s membrane, homologs of the ABC and MCF transporter families, and the presence of channels.

These data provide insight into the integration and regulation of different parts of T. cruzi metabolism, which are uniquely distributed over different compartments. This information reveals opportunities for drug development against Chagas disease caused by these parasites, for which currently no adequate treatment is available. Glycosomes are authentic members of the peroxisome family, but they are characterized by containing enzymes of the glycolytic and gluconeogenic pathways.

Fundamentals. Glycolysis occurs in the cytoplasm where one 6 carbon molecule of glucose is oxidized to generate two 3 carbon molecules of pyruvate. The fate of pyruvate depends on the presence or absence of mitochondria and oxygen in the cells. The electron transport chain is the major site of oxygen consumption and the generation of ATP in the mitochondria. In cells with mitochondria, the pyruvate is decarboxylated by pyruvate dehydrogenase complex to form Acetyl-CoA that feeds into the Tricarboxylic acid cycle and ultimately participates in ATP production.

During the absence of oxygen (anaerobic conditions) and in the cells lacking mitochondria, anaerobic glycolysis prevails. The pyruvate is reduced to lactate as NADH is reoxidized to NAD+ by lactate dehydrogenase. This process is an important source of ATP for cells that lack mitochondria, such as erythrocytes. During aerobic glycolysis, this NADH is transported by the malate aspartate shuttle or glycerol phosphate shuttle to the mitochondria, where it is reoxidized to NAD+ while it participates in the electron transport chain to produce ATP.

Cellular Level. Aerobic glycolysis is a series of reactions wherein oxygen is required to reoxidize NADH to NAD+, hence the name. This ten-step process begins with a molecule of glucose and ends up with two molecules of pyruvate.
